(For the full circumference of the pen to be filled, leaf should be at least ½” wide, sometimes more.) ● You MUST send the samples using overnight shipping, to arrive at the Lab Monday - Friday. Delayed shipping, or arrival on the weekends may result in a lack of viable tissue. You will not receive a refund if tissue samples are too deteriorated to analyze. 1 Questions? General Helpful Hints: ● For quick and easy sampling, transplant successful seedlings from cell trays into small pots (2” to 5” inches). ● The leaves MUST be young and tender. Avoid brittle, rigid or discolored tissue. ● Sampling the first set of true leaves, which grow from a seedling, can stunt the growth of the plant. The 2nd set of leaves to grow are usually ready to sample 10 to 14 days after germination. However, we recommend the 3rd set of leaves. You can sample throughout the veg cycle, as long as there is new growth large enough to fill the biopsy pen. ● Keep stakes with plants for the duration of their life cycle, for reference. Shipping: Important: Do not use USPS®. Only use FedEx® or UPS® Overnight Shipping. ● The customer is responsible for shipping samples overnight to avoid tissue deterioration. ● Hempcipher recommends FedEx Priority Overnight®, with arrival by 10:30 am the next day. The lab is only open Monday through Friday. Hempcipher is not responsible for failed results from deteriorated tissue, due to shipping methods or arrival times during Lab closure. ● Find the Express Pickup time before sampling: https://www.fedex.com/locate/ ○ Then, call the store and speak to a human to find out the cut-off time for Priority Overnight® to Idaho. Pick-up times may fluctuate due to your region, construction and natural disasters. ● Up to 5 Plates fit comfortably in a Small FedEx Express® box. Before you start: ● Check Kit for correct contents & collect additional materials. ● Find a clean workspace away from direct wind or fans. ● Ensure sample leaves are dry and free of dirt before sampling. ● Put Finger Cot on the pointer finger of non-dominant hand to test for comfort. If it is too big or small, instead wrap a piece of scotch or lab tape around the pointer finger twice, to protect the skin from cuts. Taking Samples: 1. Make sure the Plate ID number is the same for the Collection Plate and the Stakes. Look for the icon to help you quickly confirm the match (club, star, heart, etc) 2. Insert the 1st column of Stakes beside the first 8 plants to be sampled (1A, 1B, 1C, ,... 1H). . 3. To help you keep your place when sampling, position the Plate Cover over Columns 2-12, leaving Column 1 uncovered. 4. Choose a young, tender true leaf (not cotyledon), large enough for the Biopsy Pen. This should be from the newest or next to newest growth. 5. Take a sample with the Biopsy Pen. a. Wipe off any dirt or water for accurate results. b. Place cotted or taped finger underneath the leaf. c. With the other hand, place the steel cutting edge of the Biopsy Pen on the leaf, ensuring a full circle can be cut. (Do not place the steel cutting edge overlapping the edge of the leaf. ) d. Apply light pressure and twist the Biopsy Pen to cut the plant tissue. (If the sample is not cutting or staying in the Biopsy Pen, apply slightly more pressure and add an extra twist.) e. Check that the pen is filled completely with a tissue sample. f. Check the plant Stake, then place the Pen over the corresponding Well, in the 3 Questions? (Sample down the columns.) (Continued - Step 5) g. Ensure the pen is vertical, then push down on Plunger Ejector (at the top) to add the tissue to the correct Plate Well. 6. Repeat Step 5 for the remaining 7 wells in Column 1: 1B, 1C, 1D, 1E, 1F, 1G, 1H Helpful Hint: Make sure that each well only gets one sample. If a sample is placed in the wrong well, use needle-nose tweezers to remove it. 7. Move the Plate Lid over one column to the right and repeat Steps 1-6 to sample Column 2. (After you have sampled a few Columns, reorient the Lid to cover the completed columns.) 8. Repeat for the remaining Columns: 3 through 12. 9. Leave H12 empty as the Control Well to ensure accurate results. 10. When 95 Wells contain a Sample (remember to leave H12 empty), push Samples to the bottom quarter of each Well, with tweezers. 11. Peel backing off Microporous Film Sheet. Place over the Plate, adhesive side down. Press firmly across the top and wrap the edges down to seal. 12. Place a Desiccant Card (Dri-Card) on top of the Microporous Film. Place the Plate Cover on top. Rubber band the Plate to the Cover. Put the Index Card with the Plate for shipping. 4 Questions?
