TP53 gene mutations are present in more than half of all human cancers. The resulting proteins are mostly full-length with a single amino acid change and are abundantly expressed in cancer cells. Some of the mutant p53 proteins gain oncogenic functions (GOF) through which it actively contribute to the aberrant cell proliferation, increased resistance to apoptotic stimuli and ability to metastasize. Gain of function mutant p53 proteins can transcriptionally regulate the expression of a large plethora of target genes. This mainly occurs through the formation of oncogenic transcriptional competent complexes that include mutant p53 protein, known transcription factors, posttranslational modifiers and scaffold proteins. Mutant p53 protein can also transcriptionally regulate the expression of microRNAs, small non-coding RNAs that regulate gene expression at the posttranscriptional level. Each microRNA can putatively target the expression of hundred mRNAs and consequently impact on many cellular functions. Thus, gain of function mutant p53 proteins can exert their oncogenic activities through the modulation of both non-coding and coding regions of human genome. Over the past 3 decades, the regulation of p53 has been extensively studied. However, the regulation of mutant p53 remained largely unexplored. This snapshot focuses on recent discovery of mutant p53 GOF and regulation.

Recent years have seen the idea of a close association between nutrition and the modulation of cancer development/progression reinforced. An increasing amount of experimental and epidemiological evidence has been produced supporting the concept that many different bioactive components of food (e.g. polyphenols, mono- and polyunsaturated fatty acids, methyl-group donors, etc.) may be implicated in either the promotion of or the protection against carcinogenesis. At the cellular level, such compounds can have an impact on different but sometimes intertwined processes, such as growth and differentiation, DNA repair, programmed cell death, and oxidative stress. In addition, compelling evidence is starting to build up of the existence of primary epigenetic targets of dietary compounds, such as oncogenic/oncosuppressor miRNAs or DNA-modifying enzymes, which in turn impair gene expression and function. Since there is a growing interest in the study of the biochemical and molecular role played by food components and its impact on cellular processes and/or gene expressions directed towards the fine-tuning of cancer phenotypes, in this Special Issue researchers contributed with either research or review articles presenting the latest findings on the intracellular pathways and mechanisms affected by natural bioactive dietary molecules.

This book discusses the mechanical and opto-electronic design of laser spectrometers for measuring two very important atmospheric gases, namely water vapor and its isotopic ratios, and formaldehyde. For measuring water vapor, shot-noise limited sensitivity has been achieved by a careful choice of system components and data processing. For measuring formaldehyde, a selective sample modulation exploiting the Stark effect has been used to greatly improve the sensitivity.

Also, there has been an increasing number of quantitative studies investigating the connection between climate change and human societies in academia. Given this background, the Atmosphere Special Issue “Climate Change, Climatic Extremes, and Human Societies in the Past” aimed to highlight the major aspects of the climate-society nexus in ancient and recent human history. There are eight papers based on quantitative approaches to illustrate different forms of climate-society nexus in ancient, historical, and contemporary periods. Regarding ancient periods, the interconnection among climate, agriculture, and human societies is focused. Regarding historical periods, the non-linear and complex relationship between climate change and the positive checks (wars, famines, and epidemics) in historical China and pre-industrial Europe is revealed. Regarding contemporary periods, the papers focus on weather-related phenomena that significantly affect human societies. The complexity of those phenomena is also highlighted. The associated findings can help human societies to mitigate the adverse impacts of weather extremes better. This special issue contributes to the field of quantitative analysis of the climate-society nexus, both theoretically and methodologically, which could facilitate a more fruitful discussion about the climate-society nexus.
