The aetiology is complex and often multifactorial. The outcome is not simply a loss of voluntary control due to disruption of descending pathways, but also involves abnormal development of reflex and corticospinal circuitry. CP may be viewed as aberrant plasticity in response to a lesion, indeed, abnormalities in movement are subtle at first but develop subsequently. It is misleading to suppose that developmental mechanisms are self-reparative. The challenge is to understand activitydependent fine tuning of neural circuitry during normal development and to find how to promote desirable plasticity whilst limiting undesirable effects following developmental lesions. However, before proposing interventions, we have to develop our ability to predict the severity of neonatal insults.We solicited a variety of articles, including long and short reviews, original research and opinion pieces, from both basic scientists and clinicians. Social insects are among the most successful and ecologically important animals on earth. The lifestyle of these insects has fascinated humans since prehistoric times. These species evolved a caste of workers that in most cases have no progeny. Some social insects have worker sub-castes that are morphologically specialized for discrete tasks. The organization of the social insect colony has been compared to the metazoan body. Males in the order Hymenoptera (bees, ants and wasps) are haploid, a situation which results in higher relatedness between female siblings. Sociality evolved many times within the Hymenoptera, perhaps spurred in part by increased relatedness that increases inclusive fitness benefits to workers cooperating to raise their sisters and brothers rather than reproducing themselves. But epigenetic processes may also have contributed to the evolution of sociality. The Hymenoptera provide opportunities for comparative study of species ranging from solitary to highly social. A more ancient clade of social insects, the termites (infraorder Isoptera) provide an opportunity to study alternative mechanisms of caste determination and lifestyles that are aided by an array of endosymbionts. This research topic explores the use of genome sequence data and genomic techniques to help us explore how sociality evolved in insects, how epigenetic processes enable phenotypic plasticity, and the mechanisms behind whether a female will become a queen or a worker.
Wound healing plays an integral part of cellular and molecular events. Sphingosine-1-phosphate (S1P) is a lipid mediator that promotes angiogenesis and cell proliferation and attracts immune cells. S1P plays a role in skin wound healing by altering the expression of its biogenic enzyme, sphingosine kinase-1 (SphK1). SphK1 overexpression also leads to less scarring through transforming growth factor (TGF)-β1 and S1P receptor-2 (S1PR2). IFN-γ might be involved in the proliferation and maturation stages of wound healing through the regulation of neutrophilic inflammatory responses in IFN--deficient (KO) mice. Topical recombinant human (rh)-growth hormone (GH) accelerated pressure ulcer (PU) healing in non-obese diabetic/severe combined immunodeficient mice engrafted with a full-thickness human skin graft model. Matrisome properties of scaffolds that direct fibroblasts in idiopathic pulmonary fibrosis and liver regeneration are enhanced by hepatocyte-derived angiogenesis via B-cell CLL/lymphoma/nuclear factor-kappa B signaling. Mechanisms of autologous adipose-derived stem cells in some patients with human immunodeficiency virus (HIV), treated by highly active antiretroviral therapy, are elucidated. The cloning and identification of thymosin (Pa-THYs) from Periplaneta americana, the American cockroach, suggest that this molecule could be a potential drug for promoting wound repair. Maresins (MaRs) and macrophages are reviewed, focusing on the potent action of MaRs in the enhancement of M2 macrophage phenotypic profiles to possibly alleviate inflammatory pain.
In parallel with a decrease/stabilization in the use of internationally controlled drugs, the market of novel psychoactive substances (NPS) continues increasing, year after year, with the Internet playing a pivotal role in contributing to this complex scenario. At times, although misleading, the terms ‘legal highs’ or ’research chemicals’ have been used to describe these molecules. Overall, about 5% of 19–24-year-old European people have already experimented with them, because both NPS’ intense psychoactive effects and their virtual non-detectability in routine drug screenings. Although ‘novel’ typically refers to molecules that have recently become a reason of current/potential public health concern, at times, the NPS scenario may indeed focus on pharmaceutical molecules. These may have either not entered the market, or are currently prescribed, with their potential for misuse resulting from the relating high dosages/idiosyncratic ways of self-administration. A concurrent use of a range of different NPS, and/or medications, is frequently being reported and this may be a reason of further clinical complications. Given their complex pharmacodynamics, there are increasing levels of concern about the onset of acute/chronic psychopathological associated with NPS intake. This Special Issue aims at providing an overview of a range of NPS-related issues, e.g., preclinical, epidemiological, and clinical pharmacological; the medical and psychopathological consequences associated with their intake; and, finally, the analytical chemistry and forensic analysis challenges associated with the NPS phenomenon.
