The buffy coat is the fraction of an anticoagulated blood sample that contains most of the leukocytes (white blood cells) and thrombocytes (platelets) following centrifugation. After centrifugation, a thin layer forms between the clear plasma above and the red erythrocyte layer below. Composing less than 1% of the total blood volume, the buffy coat is usually buff or whitish in color — though it can appear greenish when neutrophils, which are rich in the green-pigmented enzyme myeloperoxidase, are present in large numbers. The buffy coat is widely used for DNA extraction, as leukocytes provide approximately 10 times more concentrated sources of nucleated cells than whole blood. Mammalian erythrocytes are anucleate and do not contain DNA, making buffy coat the preferred starting material for genomic research, immunology studies, infectious disease investigations, and blood disorder analysis.
